scanner. The complication rate was considerably low (11%) and comparable to the
application of this model under normal conditions.
In this experiment, the development of cytotoxic brain edema was detectable within
the first minutes following middle cerebral artery-occlusion. An increase in T2-
relaxation time, that is indicative for vasogenic brain edema, could be detected as
early as 20 minutes after induction of ischemia. This result indicated a surprisingly
early breakdown of the blood-brain barrier.
In summary, application of the macrosphere-model avoids typical side-effects of the
established suture-model. This new technique allows a closer simulation of the
clinical situation of human stroke patients and thus improves the transferability of
results derived from animal studies to human stroke. The macrosphere-model
furthermore is suitable for in-bore occlusion experiments and thus facilitates
investigation during the hyperacute phase of focal cerebral ischemia.
However, the macrosphere technique does not allow reperfusion. This circumstance
represents the most important limitation of this otherwise promising new experimental
technique.
